3D Rotating High Speed Motion

"3D Rotating High Speed Motion" is a high-speed movement which vertically rotates and moves the sample tubes in a "figure-eight" motion. This motion allows beads to impact the cells more directly for effective disruption.

Proteinase K Method vs Beads Cell Disrupter MS-100


  1. In preparation for examination, yeasts collected from the culture broth at an OD660 of 0.20 were mixed with buffer.
  2. Perform following three methods for cell disruption, two processes each;
    a. Processed without beads (as a negative control)
    b. Processed with the Proteinase K Method
    c. Processed with MS-100, using 1mm glass beads at 3,000rpm for 5 minutes
  3. After disruption, each sample was phenol extracted, ethanol precipitated and electrophoresed on a 1% agarose gel.


Genomic DNA and RNA, which were hardly extracted by using Proteinase K method, could be successfully obtained with MS-100.

Beads and Disruption Conditions

  1. φ4.8 stainless steel beads x 2
    at 3,500rpm for 90 sec x 2
  2. φ5.5 stainless steel beads x 1
    at 3,500rpm for 90 sec x 2
  3. φ3.2 stainless steel beads x 10
    at 3,500rpm for 120 sec
  4. φ4.8 stainless steel beads x 2
    at 4,000rpm for 120 sec
  5. φ5.5 stainless steel beads x 1
    at 4,000rpm for 120 sec
  6. φ5 zirconium beads x 1
    at 4,300rpm for 100 sec


MS-100 reduces the amount of low molecular weight RNA and allows recovery of high quality RNA.


  1. Add 450μl of Buffer to a 2ml tube containing a specified amount of beads.
  2. Put 80 to 90mg rice leaves into the tube.
  3. Disrupt the sample using the TOMY Beads Cell Disrupter MS-100.
  4. Perform extraction and purification according to the procedure given to the kit.
  5. After measuring A260 value of the recovered RNA solution, 5μg of total RNA of each sample was subjected to electrophoresis through a 1.2% denatured agarose gel.
